Optical-Tweezers Microscope

LUMICK C-Trap Dymo400

The LUMICK C-Trap Dymo400 is a state-of-the-art optical-tweezers microscope. It enables microscopic objects to be trapped, manipulated and fluorescently imaged, allowing single-molecule investigation of nucleic acid:protein interactions, force spectroscopy and cell mechanics.

Description

The LUMICKS C-Trap Dymo400 is a four-trap optical-tweezers microscope with integrated 3-colour confocal-microscopy and 5-channel microfluidics. It can be used to manipulate biomolecules, applying and measuring forces with pN resolution while simultaneously fluorescently imaging the sample with 3-colour scanning confocal microscopy. It is commonly used to study DNA:protein interactions by tethering individual DNA molecules between optically-trapped beads and imaging fluorescently labelled protein interacting with the DNA in real-time at single-molecule resolution. It can also be used to perform force spectroscopy to measure mechanical properties of biomolecules such as DNA, RNA and proteins. In addition measurements of cell mechanics and phase separation can be performed.

It is very user friendly and intuitive to use, requiring minimal user training compared to traditional home-built optical-tweezer microscopes. Furthermore the control software can be used to automate experiments, reducing user input and increasing reproducibility.
